I do not mind helping you through Private messages but i have found out a lot of people cant figure out my explanations.
If you PM and don't understand or to Frustrated with DB just download Teamviewer and makes both our lifes easy.
Going Almost 24hours solid since .424
I would Recommend making sure the below is installed or Repair if the option is there and Do a Complete System restart.
.Net Framework 4.5.1

AND X64 (if you have a 64bit Operating System)

C++ Redistributable 2013 x86 AND X64 (if you have a 64bit Operating System)

C++ Redistributable 2015 AND X64 (if you have a 64bit Operating System)

(( 2015 and 2013 Offer both X86 and X64 downloads once you hit the download button a menu will pop up))
Steps To Go Through:
Do a complete 100% install of Demonbuddy RELEASE .424 ( this means Not copying over your old Demonbuddy, Make a new folder and extract it to it. )
Go into the the Fresh install of Demonbuddy & Locate the Plugins Folder
You should see QuestTools and Trinity ( LEAVE THEM ALONE )
Download Adventurer .079 or the newest version out /// Extract Adventurer and Profiles Folder to the Demonbuddy PLUGINS folder

Use the Profiles Provided with Adventurer
Start Demonbuddy see if it works, if it does not come back here and explain the issue with a LOG file
