Could not get Diablo III to install on Wine. I wasn't using a full Diablo III image though, only the download and didn't get far at all. Ubuntu with VMware is a good idea.

I install all the resources through Wine so that could be the issue. I do believe using VMware and installing dependencies is a much smoother route since the client and bot will be run in a much closer to native environment.
I went back to Windows 7 to keep things simple. You're right especially in the gaming department, a lot of 3D applications just don't handle Wine well, if at all. Even a VM on Ubuntu can be rocky, DirectX being the main concern.
